YIJI Protein Sample Resin Method Endotoxin Kit Instructions

YIJI protein sample resin detoxification kit

The main purpose

YIJI Protein Sample Resin De-endotoxin reagent is an authoritative and classical technical method aimed at removing bacterial endotoxin from samples by filtration and adsorption by lipopolysaccharide affinity resin. The technology has been carefully developed and successfully tested. It is suitable for the preparation of various bacterial protein purified samples. The product can be used in a variety of subsequent animal and cell biology operations. The product is ready to use, stable performance, convenient operation and satisfactory cleaning efficiency.

technical background

Endotoxin is a component of Lipopolysaccharides (LPS), a cell membrane of Gram-negative bacteria. During the purification of bacterial proteins, including fusion proteins, bacterial lysis releases endotoxin, accompanied by proteins that severely interfere with subsequent sensitive animal and eukaryotic cell experiments. YIJI protein sample resin detoxification technology incorporates lipopolysaccharide-specific affinity resin, namely polylysine coated and functionalized cation exchange groups, ultra-highly crosslinked polystyrene polymerization. Hyper-crosslinked polystyrene polymers are sufficiently effective to filter and remove endotoxin in the sample by 10 μg/ml, ie 2500 to 5000 EU/ml. Non-specific protein binding by chromatography, loss of immunological activity, and partial endotoxin failure and expensive disadvantages are avoided.

Product manual 1 copy

storage method

Stored in a 4 ° C refrigerator, effective for June

User-supplied

1.5 ml centrifuge tube: container for sample handling

Mini Tabletop Centrifuge: for Precipitating Affinity Resins

Vortex oscillator: used to mix reactants

Flat shaker: used to mix reactants

Experimental procedure

Before the start of the experiment, the sample of the protein to be tested was taken out from the -70 ° C refrigerator and thawed in an ice bath. Then do the following.

  • Add 1 ml of protein suspension (1 to 5 mg of total protein) to a 1.5 ml centrifuge tube
  • Carefully remove 150 μl of YIJI Affinity Reagent (Reagent A) into a 1.5 mL centrifuge tube
  • Cover the lid and vortex for 15 seconds
  • Place it on a flat shaker and incubate at 4 ° C for 3 to 16 hours at 100 RPM
  • Place in a mini tabletop centrifuge for 5 minutes at a speed of 500g (or 2000RPM, eg eppendorf 5415D)
  • Carefully remove the supernatant into a new 1.5 ml centrifuge tube to avoid black granules
  • Immediately put in a -70 ° C refrigerator

Precautions

  • This product is 20 operations
  • This product uses a special resin method
  • Use a 1 ml tip to cut off the tip of the tip for removal of YIJI Affinity (Reagent A)
  • YIJI Affinity Liquid (Reagent A) cannot be reused
  • The average endotoxin content of 100 ml of bacteria can be as high as 15,000 EU (30 μg), between 5000 and 50000 EU
  • The degree of endotoxin removal depends on the endotoxin content in the sample, the nature of the sample (bacterial source, fusion protein properties) and solution conditions (pH, salt ionic strength, etc.), which can be completely removed by multiple treatments; Ideal, it is recommended to use YIJI protein sample liquid phase detoxification kit-YIJI30046.2
  • The product treated with this product contains 90% protein content and 100% immunogenicity.
  • The company provides a series of endotoxin treatment reagent products

Quality Standard

  • This product has been certified to be stable.
  • This product has been identified to be significantly endotoxin
