Fudan University bred transgenic zebrafish that can monitor estrogen contamination

A magical “transgenic zebrafish” that can visually monitor environmental estrogen contamination was successfully cultivated at Fudan University in Shanghai and accepted by experts. The task force of the key project of the Natural Science Foundation of Shanghai, led by Professor Song Houyan and Professor Zhong Tao, at the Key Laboratory of Molecular Medicine of the Ministry of Education, Fudan University, completed the work.

Environmental estrogens are widely found in water, soil, and food. Through bio-enrichment, the concentration of environmental estrogens in living organisms is approximately tens of times higher than that in the environment. Environmental estrogens severely interfere with the development of gonads in wildlife, leading to individual reproductive dysfunction and reduced population. As long as a very small amount of environmental estrogen enters the body, it will have an adverse effect on male reproductive function. Over the past 50 years, the global men's semen quality has decreased significantly, resulting in an increase in infertility rates and congenital malformations. The increase in the incidence of testicular cancer and prostate cancer may also be related to environmental estrogen contamination.

Three years ago, the research team introduced and built a large-scale, automated model of biological zebrafish breeding technology platform and a clear genetically pure zebrafish species fish from the United States. After a long period of arduous and meticulous work, the zebrafish vtg promoter, which is sensitive to estrogens, was cloned and identified. An eukaryotic expression plasmid for the gfp gene regulated by this promoter was constructed and microinjected into the eukaryotic gfp gene. The expression plasmid was transferred into the zebrafish fertilized egg. After a large number of screenings, the F1 transgenic zebrafish, which is sensitive to estrogens, was finally obtained.

The fish can visually, sensitively, specifically, conveniently, and quickly show estrogenic pollution in the water environment. Even if the level of environmental estrogen contamination in the water reaches only a very small extent, the liver of the transgenic zebrafish emits green fluorescence.

Experts believe that genetically modified zebrafish is very convenient and rapid for the detection of estrogen in aquatic environments, and has important application and theoretical research value for environmental protection, reproductive physiology, and reproductive pathology.
