Application of gel imaging system and computer software to bioengineering research

1 image intake
The computer gel imaging system SX100 (Shanghai Four Star Biotechnology Industrial Co., Ltd.) uses digital photography to directly input the images taken into the computer system. The sample is clear and sized by adjusting the zoom aperture, zoom factor and focal length under UV light in a black box. After image acquisition is obtained, brightness adjustment and image optimization are performed by "brightness" and "optimization" of the image processing menu in BioImageV2.0 software to reduce the image noise floor. Generally, after this processing, clear condensation can be obtained. The glue picture, while adding text information to the image for proper annotation, can be printed with a thermal printer or a high-quality photo-level inkjet printer (such as EPSON's PHOTO series) that is commonly used today to obtain photo prints that meet the published requirements of the article.

2 molecular weight quantification
For the commonly used DNA film, the molecular weight quantification function in BioImage is used to automatically generate a fitting curve by annotating the known molecular weight of the DNA Marker band on the gel, and measuring the molecular weight of the unknown band by it. The results obtained by this method are much more accurate than the visual observations. For example, the molecular weight is judged by its function when the restriction endonuclease digestion verification is performed after the expression vector pBLII is constructed.

3 Density quantification
A commonly used method for determining the concentration of DNA (deoxyribonucleic acid) and RNA (ribonucleic acid) is the ultraviolet absorption method, but it can only measure the total nucleotide concentration in the sample, and cannot distinguish the concentration of each length fragment. However, using the gel imaging system and BioImage software, after adjusting the threshold and the integral frame and performing density calibration on a standard strip on the DNA film, it is convenient to click other unknown strips according to the standard strip. Absolute optical density values ​​give their DNA content. It is more convenient than the ultraviolet absorption method, and the content of fragments of different lengths can be measured, and the obtained result is sufficient for the accuracy required for molecular biological operation. It is worth noting that this method is also applicable to the determination of the concentration of the PA GE protein strip.

4 density scanning
In molecular biology and bioengineering research, the most commonly used is the calculation of the percentage of protein expression products in the total bacterial protein. The traditional approach is to use a dedicated density scan, but this can be done using bioanalytical software in conjunction with a scanner that is routinely equipped in the lab or a gel that is directly illuminated with white light.

Taking BioImage V2.0 as an example, the density scan command is selected, and the image of the lane representing a certain microbial strain is framed by a dotted line, and a longitudinal scan curve of the region is obtained. Generally, the default automatic integration option is selected to automatically calculate the percentage of each peak area corresponding to each tape relative to the entire curve. You can also manually select the integral area by manual integration. For some bands, the background is weak. Higher spectra can be baseline adjusted for better experimental results and within the tolerances. The figure shows the expression of the constructed pBL II vector and the SDS-PA GE electrophoresis detection and the fourth lane scanning protein expression as a percentage of total protein expression, the result is about 25%.



5 PCR (Polymerase Chain Reaction)

PCR quantification mainly refers to the fact that if the amplified bands are not one when doing PCR experiments, the relative percentage of each band can be calculated by using this command. This function is similar to the density scan, but the actual principle is not the same. PCR quantification does not generate a longitudinal scan of the selected region and integrates it, but instead quantifies the relative density of the selected bands and calculates their percentage of the sum. Figure 4 is a graph showing the content of each fragment in the first lane PCR result on the left.

6 Summary and recommendations
This article describes the functions of gel imaging systems that are encountered almost every day in molecular biology and bioengineering research. Because of the combination of biological processing software, it can cover various fields of daily research in modern biological sciences and can replace them.
